Figure Top: Shows a contig overview of two sequence reads from the 3' end of a cDNA clone. The Poly T-primed sequence reads much further into the ORF than the 3' vector primer sequence, and its sequence quality is far more accurate for EST analysis and selecting for full-length clones.

Figure Bottom: Top read was obtained with a Poly T primer. The bottom read was obtained using the 3' vector primer which produces "mixed/low-quality" sequence after passing through the Poly A (reverse-comp) tail.
