cDNA Library Construction

Beginning with a minimum of 100µg of total RNA, 1µg of poly(A+) RNA, or 100mg of tissue, SeqWright will construct your customized cDNA library. Average turnaround time is 10-12 weeks. Service includes:

  • Full-length cDNA preparation
  • cDNA normalization (typically using Cot10, Cot50 or Cot500)
  • Ligation of normalized cDNA into a selected vector system
  • Transformation of ligate into E. coli.
  • Plating and arraying of 96 clones
  • 96-well culturing and purification
  • Single pass confirmation sequencing
  • Normalization efficiency analysis using PCR (optional, on request)
  • Subtracted cDNA libraries are also available
